I understand what you are asking and I do not believe that you can. However, I am speaking from a 9.2 standpoint. There may be different functionality in the later versions.

Yes, use the "Table to Domain" tool in ArcToolBox "Data Management Tools; Domains".

fairly straighforward as long as you get the table formatting clean. I usually import the table into a gdb table first to make sure; but excel files do work as well as text etc.
