I have two servers one Arcgis server and other Sde server with MS-Sql. I have create arcsde instance on sde server.
ON arcgis server am able to create spatial connection using sde instance. but not able to create direct connection

Are you starting the instances that you're using through cloud builder? Cloud builder will setup the connection for you automatically.
